In cell culture, serum is an essential nutrient that enhances adhesion factors, growth factors, binding proteins, etc., for cell growth. When using serum, we will be involved in the operation of serum loading, so how should it be packed into PETG serum bottles?
1, defrost
Remove the serum from the refrigerator at -20 degrees Celsius and freeze it at room temperature (or in tap water) (about 30 minutes to 2 hours, or put it in the refrigerator at 4 degrees Celsius overnight; If it is not inactivated immediately after thawing, it can be temporarily stored in a 4 degree refrigerator).
2, inactivated
Water bath at 56 ° C for 30 minutes and shake evenly at any time. Remove and cool immediately on ice. Allow to cool to room temperature (1-3 hours). In the process of thermal inactivation, the occurrence of precipitation can be reduced by periodic shaking.
3, packing
Transfer to the sterile room, separate the serum into 50-100ml PETG serum bottles in the ultra-clean table, seal them, and store at -20℃ for later use. In the packaging should pay attention to: in advance to gently shake the serum for several weeks, mix; When blowing out the serum with the suction tube, be careful: do not blow bubbles, the serum is very sticky and easy to bubble. If bubbles are produced, run them over the flame of an alcohol lamp.
The above are the specific operation steps of serum packaging. Please do not put your hands above the open bottle mouth. The packaging speed should be fast to avoid sedimentation bacteria falling into the bottle mouth of PETG serum bottle.
