When culturing Jurkat cells in erlenmeyer shake flasks, RPMI1640 medium, 10% FBS is required; the temperature is controlled at 37°C, 5% carbon dioxide, PH value 7.2-7.4, aseptic constant temperature culture. Wipe and disinfect with 75% alcohol before transferring to the cell ultra-clean bench, take out the cell cryovial from the liquid nitrogen tank, immediately place it in a 37°C water bath and shake the cell cryotube quickly to thaw it quickly. Then, after centrifugation, pipetting and mixing, etc., it was placed in a cell incubator for cultivation.
Cells are particularly sensitive to the environment. When culturing Jurkat cells in erlenmeyer cell shake flasks, personal hygiene should be done well, sterile reagents should be used, and aseptic operation principles should be followed to avoid introducing bacteria and affecting cell growth.
